چکیده

The product of the Drosophila extradenticle gene interacts cooperatively with homeodomain proteins encoded by homeotic selector genes, and may account in part for their distinct regulatory properties.

Expression and Purification of Homeodomain

Homeobox genes encode transcription factors which play important roles in the developmental processes of many multicellular organisms. TGIFLX/Y (TGIFLX and TGIFLY) are members of the homeobox superfamily of genes. Their expressions are specifically detected in the human adult testis but their functions are remained to be investigated. In this investigation we cloned full length of TGIFLY cDNA a...

The homeodomain of PDX-1 mediates multiple protein-protein interactions in the formation of a transcriptional activation complex on the insulin promoter.

Activation of insulin gene transcription specifically in the pancreatic beta cells depends on multiple nuclear proteins that interact with each other and with sequences on the insulin gene promoter to build a transcriptional activation complex. The homeodomain protein PDX-1 exemplifies such interactions by binding to the A3/4 region of the rat insulin I promoter and activating insulin gene tran...

Drosophila homeodomain protein REPO controls glial differentiation by cooperating with ETS and BTB transcription factors.

In Drosophila, cell-fate determination of all neuroectoderm-derived glial cells depends on the transcription factor Glial cells missing (GCM), which serves as a binary switch between the neuronal and glial cell fates. Because the expression of GCM is restricted to the early phase of glial development, other factors must be responsible for the terminal differentiation of glial cells. Expression ...

An Oct-1 binding site mediates activation of the gata2 promoter by BMP signaling

The gata2 gene encodes a transcription factor implicated in regulating early patterning of ectoderm and mesoderm, and later in numerous cell-specific gene expression programs. Activation of the gata2 gene during embryogenesis is dependent on the bone morphogenetic protein (BMP) signaling pathway, but the mechanism for how signaling controls gene activity has not been defined. We developed an as...

Both Pbx1 and E2A-Pbx1 bind the DNA motif ATCAATCAA cooperatively with the products of multiple murine Hox genes, some of which are themselves oncogenes.

E2A-PBX1 is the oncogene produced at the t(1;19) chromosomal breakpoint of pediatric pre-B-cell leukemia. Expression of E2A-Pbx1 induces fibroblast transformation and myeloid and T-cell leukemia in mice and arrests differentiation of granulocyte macrophage colony-stimulating factor-dependent myeloblasts in cultured marrow. Recently, the Drosophila melanogaster protein Exd, which is highly relat...
